Chisako Kakehi, who benefited to the tune of 800 million yen ($6.8 million) over the past 20 years, according to Japanese media, denies any wrongdoing.
"I didn't kill him. I don't know how to kill ..." she told police, according to CNN affiliate TV Asahi.
But an autopsy report on Iso Kakehi, her fourth husband to die, found cyanide in his stomach and blood.
The 75-year old died in December, less than two months after their wedding. It triggered a police investigation into the case, which culminated in Kakehi's arrest in Kyoto on Wednesday.

It's the latest example of the complications Bush has faced on the immigration issue as he considers a 2016 presidential run. Bush has long drawn criticism from conservatives for his more moderate positions on reform, including his support for a pathway to citizenship for those here illegally.
His Friday statement on Obama's executive action to protect nearly 5 million immigrants from deportation was nuanced, taking aim only at the tactic, not the ultimate effect.
"President Obama's ill-advised unilateral action on illegal immigration undermines all efforts to forge a permanent solution to this crisis. Action must come in the form of bipartisan comprehensive reform passed through Congress," Bush said.
He later tweeted that the move was "an abuse of power."

Ever since winning last year’s Arab Idol, the Palestinian has been touring the world, performing a variety of covers ranging from Palestinian and Lebanese folk songs to Khaleeji pop.
With his self-titled debut album out last month through MBC’s Platinum Records, Abu Dhabi fans are now set to be among the first in the region to finally hear some of Assaf’s original tunes when he headlines at du Arena on Thursday in an Arab music spectacular that also features Egypt’s Tamer Hosny, Lebanon’s Carole Samaha and the UAE’s Fayez Al Saeed.
With 13 new songs to pick from, Assaf says he is raring to go on stage.
“This is a big and important step in my career, because everyone was expecting the album, especially after the huge steps that I have accomplished so far, thanks to the fans,” he says. “By the way, allow me to say that I have always considered the patriotic Palestinian songs that I have performed on stage as if they were songs of my own, and that’s the way every Palestinian feels.”

But the lack of ruthlessness and the naivety which has plagued Arsenal in recent years cost them dear at the Emirates Stadium again as a second-half own goal by Kieran Gibbs and Wayne Rooney's breakaway finish gave United the points.
"We have not dominated many games against United like we did today but we were not efficient enough and made defensive mistakes," Wenger told a news conference.
